Independent simultaneous 3D holographic optical trapping and confocal imaging

We present a unique system, combining an upright confocal microscope and an inverted microscope through which a set of Holographic Optical Tweezers (HOTs) are inserted. This combination allows us to use the confocal microscope to image the structures built by the HOTs in three dimensions. The setup includes reflected and transmitted bright field as well as fluorescence imaging. We demonstrate our ability to accurately trap and image particles in all three dimensions and correct for deviations from the desired locations using adaptive optics.
